    Efficiency of time series homogenization: method comparison with 12 monthly temperature test datasets

    The aim of time series homogenization is to remove nonclimatic effects, such as changes in station location, instrumentation, observation practices, and so on, from observed data. Statistical homogenization usually reduces the nonclimatic effects but does not remove them completely. In the Spanish ‘‘MULTITEST’’ project, the efficiencies of automatic homogenization methods were tested on large benchmark datasets of a wide range of statistical properties. In this study, test results for nine versions, based on five homogenization methods—the adapted Caussinus-Mestre algorithm for the homogenization of networks of climatic time series (ACMANT), ‘‘Climatol,’’ multiple analysis of series for homogenization (MASH), the pairwise homogenization algorithm (PHA), and ‘‘RHtests’’—are presented and evaluated. The tests were executed with 12 synthetic/surrogate monthly temperature test datasets containing 100–500 networks with 5–40 time series in each. Residual centered root-mean-square errors and residual trend biases were calculated both for individual station series and for network mean series. The results show that a larger fraction of the nonclimatic biases can be removed from station series than from network-mean series. The largest error reduction is found for the long-term linear trends of individual time series in datasets with a high signal-to-noise ratio (SNR), where the mean residual error is only 14%–36% of the raw data error. When the SNR is low, most of the results still indicate error reductions, although with smaller ratios than for large SNR. In general, ACMANT gave the most accurate homogenization results. In the accuracy of individual time series ACMANT is closely followed by Climatol, and for the accurate calculation of mean climatic trends over large geographical regions both PHA and ACMANT are recommended.This research was funded by the Spanish MULTITESTproject (Ministry of Economics and Competitiveness, CGL2014-52901-P)

    Ambient-aware continuous care through semantic context dissemination

    Background: The ultimate ambient-intelligent care room contains numerous sensors and devices to monitor the patient, sense and adjust the environment and support the staff. This sensor-based approach results in a large amount of data, which can be processed by current and future applications, e. g., task management and alerting systems. Today, nurses are responsible for coordinating all these applications and supplied information, which reduces the added value and slows down the adoption rate. The aim of the presented research is the design of a pervasive and scalable framework that is able to optimize continuous care processes by intelligently reasoning on the large amount of heterogeneous care data. Methods: The developed Ontology-based Care Platform (OCarePlatform) consists of modular components that perform a specific reasoning task. Consequently, they can easily be replicated and distributed. Complex reasoning is achieved by combining the results of different components. To ensure that the components only receive information, which is of interest to them at that time, they are able to dynamically generate and register filter rules with a Semantic Communication Bus (SCB). This SCB semantically filters all the heterogeneous care data according to the registered rules by using a continuous care ontology. The SCB can be distributed and a cache can be employed to ensure scalability. Results: A prototype implementation is presented consisting of a new-generation nurse call system supported by a localization and a home automation component. The amount of data that is filtered and the performance of the SCB are evaluated by testing the prototype in a living lab. The delay introduced by processing the filter rules is negligible when 10 or fewer rules are registered. Conclusions: The OCarePlatform allows disseminating relevant care data for the different applications and additionally supports composing complex applications from a set of smaller independent components. This way, the platform significantly reduces the amount of information that needs to be processed by the nurses. The delay resulting from processing the filter rules is linear in the amount of rules. Distributed deployment of the SCB and using a cache allows further improvement of these performance results

    Multicenter Phase 2 Trial of Sirolimus for Tuberous Sclerosis: Kidney Angiomyolipomas and Other Tumors Regress and VEGF- D Levels Decrease

    Tuberous sclerosis (TSC) related tumors are characterized by constitutively activated mTOR signaling due to mutations in TSC1 or TSC2.We completed a phase 2 multicenter trial to evaluate the efficacy and tolerability of the mTOR inhibitor, sirolimus, for the treatment of kidney angiomyolipomas.36 adults with TSC or TSC/LAM were enrolled and started on daily sirolimus. The overall response rate was 44.4% (95% confidence intervals [CI] 28 to 61); 16/36 had a partial response. The remainder had stable disease (47.2%, 17/36), or were unevaluable (8.3%, 3/36). The mean decrease in kidney tumor size (sum of the longest diameters [sum LD]) was 29.9% (95% CI, 22 to 37; n = 28 at week 52). Drug related grade 1-2 toxicities that occurred with a frequency of >20% included: stomatitis, hypertriglyceridemia, hypercholesterolemia, bone marrow suppression (anemia, mild neutropenia, leucopenia), proteinuria, and joint pain. There were three drug related grade 3 events: lymphopenia, headache, weight gain. Kidney angiomyolipomas regrew when sirolimus was discontinued but responses tended to persist if treatment was continued after week 52. We observed regression of brain tumors (SEGAs) in 7/11 cases (26% mean decrease in diameter), regression of liver angiomyolipomas in 4/5 cases (32.1% mean decrease in longest diameter), subjective improvement in facial angiofibromas in 57%, and stable lung function in women with TSC/LAM (n = 15). A correlative biomarker study showed that serum VEGF-D levels are elevated at baseline, decrease with sirolimus treatment, and correlate with kidney angiomyolipoma size (Spearman correlation coefficient 0.54, p = 0.001, at baseline).Sirolimus treatment for 52 weeks induced regression of kidney angiomyolipomas, SEGAs, and liver angiomyolipomas. Serum VEGF-D may be a useful biomarker for monitoring kidney angiomyolipoma size. Future studies are needed to determine benefits and risks of longer duration treatment in adults and children with TSC.Clinicaltrials.gov NCT00126672

    The NOMAD experiment is a short base-line search for νμντ\nu_{\mu}\rightarrow \nu_{\tau} oscillations in the CERN neutrino beam. The ντ\nu_{\tau}'s are searched for through their charged-current interactions followed by the observation of the resulting τ\tau^{-} through its electronic, muonic or hadronic decays. These decays are recognized using kinematical criteria necessitating the use of a light target which enables the reconstruction of individual particles produced in the neutrino interactions. This paper describes the various components of the NOMAD detector: the target and muon drift chambers, the electromagnetic and hadronic calorimeters, the preshower and transition radiation detectors, and the veto and trigger scintillation counters. The beam and data acquisition system are also described. The quality of the reconstruction of individual particles is demonstrated through the ability of NOMAD to observe Ks0^0_{\rm s}'s, Λ0\Lambda^0's and π0\pi^0's. Finally, the observation of τ\tau^{-} through its electronic decay being one of the most promising channels in the search, the identification of electrons in NOMAD is discussed
